Quercetin-induced apoptosis is negatively regulated by inactive glycogen synthase kinase-3βvia suppression of JNK-p53 pathway in lung epithelial cells

Abstract
Quercetin, a ubiquitous bioactive plant flavonoid, has been shown to exert anti-proliferative and apoptotic activities in some cells. Its apoptotic activity has been reported to be associated with inhibition of enzymes involved in proliferation, such as PKC, PI-3K, Na+K+ ATPase, and c-Jun N-terminal kinase (JNK) and modulation of the expression of apoptosis-associated proteins, such as p53, p21WAF/CIP1, Bax, and Bad. GSK-3β is an important modulator of apoptosis. However, the regulation of GSK-3β on the quercetin-induced apoptosis remains unclear. This study was undertaken to elucidate the role of GSK-3β in the quercetin-treated lung epithelial cells. Quercetin decreased cell viability via apoptosis dose and time-depedently. Quercetin activated JNK and increased c-Jun expression. At the same time, quercetin increased the levels of p53-dependent Bax expression. Blocking of JNK activiation by overexpression of dominant negative JNK1 suppressed apoptosis by quercetin via inhibition of caspase-3 activation and reduction of p53 and Bax expression. Moreover, GSK-3β was inactivated by uercetin. Its blocking by overexpression of constitutively active GSK-3β enhanced quercetin-induced JNK activation. In contrast, overexpression of dominant negative GSK-3β suppressed both JNK activation and apoptosis by quercetin. Taken together, our results suggest that JNK-p53 pathway is involved in quercetin-induced apoptosis and simultaneous inactivation of GSK-3β by quercetin can attenuate apoptosis in lung epithelial cells.
